ABSTRACT

Objective:To investigate the changes of serum Golgi protein 73 (GP73) expression and its clinical significance in breast cancer patients before and after radiotherapy.Methods:A total of 135 patients with primary breast cancer who were diagnosed and treated in Qingdao Hospital Affiliated to Shandong First Medical University Hospital from Aug. 2016 to Dec. 2017 were selected and received standard radiotherapy after surgery. Enzyme-linked immunosorbent assay (ELISA) was used to determine the content of GP73 in the serum of patients before and after radiotherapy, and the differences in the expression levels of GP73 before and after radiotherapy in patients with different molecular phenotypes, tumor stages, and pathological types were compared. Association between the expression of serum GP73 and clinical outcome before and after radiotherapy was analyzed.Results:The expression of GP73 in breast cancer patients was (117.69±33.57) mg/L before radiotherapy and (101.88±30.92) mg/L after radiotherapy, and the difference was statistically significant ( t=4.025, P<0.001) . Different molecular phenotypes were stratified and found that the expression of serum GP73 in patients with luminal A, luminal B, HER-2 overexpression, and triple negative after radiotherapy decreased compared with those before radiotherapy, but only the HER-2 overexpression type had statistically significant difference between before and after radiotherapy ( P<0.05) . Stratification according to different tumor stages showed that the expression of serum GP73 in patients with stage I, II, III and IV after radiotherapy was lower than that before radiotherapy, but only the patients with stage II, stage III, and stage IV were compared before and after radiotherapy, and the difference was statistically significant ( P<0.05) . Stratification according to different pathological types showed that the expression of serum GP73 in patients with invasive ductal carcinoma before and after radiotherapy was significantly lower in patients with invasive lobular carcinoma ( P<0.05) . In addition, the 1-year survival rate of the descending group was significantly higher than that of the ascending group, while the local recurrence rate and the occurrence of distant metastasis were significantly lower than those of the ascending group, and the difference was statistically significant ( P<0.05) . Conclusions:The expression level of serum GP73 in breast cancer patients after radiotherapy is significantly higher than that before radiotherapy, and it has a certain relationship with molecular phenotype, tumor stage, and pathological type. At the same time, the increase in serum GP73 expression after radiotherapy may be detrimental to the clinical outcome of patients.

ABSTRACT

Objective:To investigate the effects of music training and art training on the audiovisual integration of letters-speech sounds in college students.Method:Visual letters and auditory speech sounds were used to conduct a discrimination task test in music students, art students and ordinary college students ( n=30 for each group). SPSS17.0 was used to conduct ANOVA and Race model analysis on reaction time and hit rate. Results:(1) Reaction time: the results showed that the interaction between group and stimulus type was significant( F=7.89, P<0.01). The simple effect analysis found that all the participants reacted faster to consistent audiovisual(letters B and speech B) stimuli in comparison to unimodal stimuli, and the redundant signal effect occured.(2) Race model analysis showed that the time window of audiovisual integration were 120-130 ms in the music group, 130-190 ms in the art group, and 120-170 ms in the general group( P<0.01). The positive area (pAUC) under the Race model curve of the music group(18.95) was significantly smaller than that of the art group(159.43) and the general group(125.01)( P<0.01), and there was no significant difference between the art group and the general group( P=0.13). Conclusion:Music training has a regulating effect on the integration of letters and speech sounds, while art training has no significant effect on it.

Conclusion The1.5T MRIDEAL-IQsequencecanbeusedtoevaluatetheliverreserveunctionoflivercirrhosispatientsaccording ffatfractionandR2?value,especiallyfortheChild-PughCcirrhosis patientswithhighsensitivityandspecificity.

ABSTRACT

Taking Dafeng county of Jiangsu province as a pilot, this paper analyzed the mechanism of the family physicians′contracted service for achieving the hierarchical medical system. Such a system is designed to promote the contracted service of family physicians, comprising health management, convenient medical treatment, opinion leader, IT-based power, and capability enhancement. Authors of the paper rounded up data of the sample township from 2015 to 2017, and the practical effect of the family physicians′contracted service in promoting the hierarchical medical system. Their recommendations include:currently focusing on targeted groups and population of chronic diseases; elevating the contracting rate of pregnant women and children; motivating countywide medical communities in the promotion of the family physicians′contracted service, for joint efforts in developing the hierarchical medical system; leveraging the showcase of Dafeng for achieving the hierarchical medical system fitting local needs.

ABSTRACT

Background and purpose: Factor that binds to the inducer of short transcripts of human immuno-deficiency virus-1 (FBI-1) in a variety of malignant tumors showed high expression levels, which may be closely related to tumor proliferation and differentiation, angiogenesis, metastasis, but its relationship with breast cancer has not been fully elucidated. The purpose of this study was to investigate the expression of FBI-1 in breast cancer cells, and to study the effect of FBI-1 gene expression on the proliferation of breast cancer cells and its possible mechanism. Methods:Real-time fluorescent quantitative polymerase chain reaction (RTFQ-PCR) and Western blot analysis were applied to detect FBI-1 expression in normal human mammary epithelial cell line MCF-10A and breast cancer cell MCF-7. RNA interference method was used to down-regulate FBI-1 expression in MCF-7 cells. The cell proliferation was measured by CCK-8 kit and colony formation assay. RTFQ-PCR and Western blot were used to detect the expression of FBI-1 and NF-κBp65 in MCF-7 cells before and after the interference of FBI-1 expression. Results: The expression of FBI-1 was higher in breast cancer cells than that in normal human mammary epithelial cells (P<0.05). The effects of FBI-1 down-regulation inhibited proliferation in MCF-7 cells (P<0.05). At the same time, after inhibition of FBI-1, the NF-κBp65 mRNA and protein expression levels were significantly decreased (P<0.05). Conclusion: FBI-1 is highly expressed in breast cancer cells. Down-regulated FBI-1 expression can inhibit the proliferation of breast cancer cells,and its mechanism may be related to the inhibition of NF-κB signaling pathway.

ABSTRACT

Objective To explore the effects and mechanism of triptolide on proliferation and apoptosis of breast cancer MCF-7 cells.Methods MCF-7 cells were treated by different concentrations of triptolide.CCK-8 as-say was employed to detect the cell proliferation. The morphological changes were observed by an inverted micro-scope.The apoptosis rate was detected by flow cytometry.Expressions of Bcl-2,Bax,Survivin and Caspase-3 were measured by qRT-PCR and Western blot. Results Triptolide inhibited the proliferation of MCF-7 cells in a dose and time-dependent manner at a suitable range.Triptolide induced morphological changes and apoptosis.Triptolide also down-regulated Bcl-2 and Survivin expressions and up-regulated Bax and Caspase-3 expressions. Conclu-sions Triptolide inhibits proliferation and induces apoptosis of MCF-7 cells,and its mechanism may be related to down-regulation of Bcl-2 and Survivin expressions and up-regulation of Bax and Caspase-3 expressions.

ABSTRACT

PurposeTo evaluate the significance of ultrasonic elastography strain ratio, MRI and the combination of both in diagnosis of breast tumor.Materials and MethodsFifty-four cases with single breast tumor underwent preoperative ultrasound elasticity imaging and MRI. Accuracy of ultrasound elastography strain rate ratio (SRR) of the tumor and surrounding normal breast tissue was measured by quantitative ultrasound elastography, and its combination with MRI were analyzed. ResultsThere was signiifcant differences on SRR between the benign group and the malignant group (2.24±1.28vs 4.96±1.73, t=2.648,P0.05). Combined both had higher diagnosis accuracy when compared with SR and MRI separately (χ2=6.000 and 3.967,P<0.05).Conclusion Ultrasonic elastography strain ratio is accurate and objective in differentiating benign from malignant breast tumors. It is a valuable quantitative index in clinical practice. Moreover, SRR combined with MRI can reduce the misdiagnosis rate.

ABSTRACT

Objective: To analyze the current situation and effect of vertical integrated of rural health services in Dafeng County. Method: Policy documents and data are collected and interviews are implemented for studying the integrated situation. Results: The vertical integration in Dafeng County is virtual joint form with forming 6 city-level medical volunteer service teams and 52 health management teams. With the imple-mentation of fully integrated management, signing service with rural doctor mode is carried out, which pro-mote the establishment of rural unified service standards and regulations. Using health card of residents which promote the regional shared of treatment information. Conclusions: Vertically integrated mode had connection in health management teams, supported by fully integration, developed by signing service with rural doctor and carried by residents’ health card, which could provide reference for implementing vertical integration of rural health service. But strategies needed to be strengthened in improving health services’ ability of township hospitals, clarifying interest relationship as well as integrating health service regulations and strengthen the health information system for sharing and docking.

Autophagy is a vacuolar process of cytoplasmic degradation by lysosomes which ubiquitously occur in all eukaryotic cells.Heightened autophagy is a mechanism of resistance for cancer cells faced with metabolic and therapeutic stress,revealing opportunities for exploitation as a therapeutic target in cancer treament.This article reviews the mechanism of autophagy in cancer and its role for cancer treatment.

ABSTRACT

Objective To identify the role of p53 in the induction of growth arrest DNA damage-inducible gene 45β (GADD45β) in HCC cells by Oxaliplatin.Methods A Hep3B+p53 clone was established by transfection of the full-length p53 sequence to Hep3B.Following oxaliplatin administration,quantitative real-time PCR was employed to validate the expression changes of GADD45β.pGL3 basic luciferase plasmids including promoter fragments were synthesized in vitro and transfected into cells.The effects on promoter activity,cell growth and the cleavage of Caspase-3 were further focused on.Results Hep3B+p53 expressed p53 protein stably.The transfection of p553 enhanced the induction of GADD45β in Hep3B by Oxaliplatin.The promoter activity of fragments constructed NF-κB and E2F-1 binding sites was induced about 1.5 and 0.8 folds by transfection of p53.The colony formation and DNA syntheses were inhibited apparently in Hep3B+p53 with p53 by Oxaliplatin (30.41% and 75.60% by 100 μmol/L Oxaliplatin,respectively).Moreover,p53 transfection triggered cleavage of Caspase-3 more rapidly.Conclusion p53 played a role in the induction of GADD45β in Hep3B by Oxaliplatin.

ABSTRACT

OBJECTIVE@#To observe the effects of N-acetyl-L-cysteine (L-NAC) protect hair cells in the rat cochlea from injury of exposure to styrene.@*METHOD@#Seventeen adult Long Evans rats were used in present study. The animals were randomly assigned into test group (n=9) and control group (n=8). The animals were exposed to styrene by gavage at 400 mg/kg (2 g styrene was mixed with 1 ml olive oil). Test group animals received styrene exposure plus L-NAC 325 mg/kg (L-NAC was dissolved in physiological saline solution) by intraperitoneal injection. Treatment was performed once a day, 5 days per week for 3 weeks. Control group animals received the same volume of saline injection on an identical time schedule used for the test group. The auditory brainstem response (ABR) thresholds of both ears elicited with clicks were measured before and at the end of the 3-week styrene or styrene plus L-NAC treatment. After hearing was re-assessed, animals were sacrificed and cochleae were quickly removed from the skull. Following fixation, whole specimens comprising the basilar membrane with Corti's organ were separated from the modiolus. The organs of Corti were stained with propidium iodide (PI) and the TUNEL assay to visualize the morphologic viability of hair cell nuclei, FITC-labeled phalloidin, a F-actin intercalating fluorescent probe used to visualize the morphologic viability of cuticular plate and the stereocilia in the hair cells. Each organ of Corti was thoroughly examined using fluorescence microscopy. The numbers of damaged OHCs (apoptotic, necrotic and missing OHCs) were documented.@*RESULT@#There was a statistically significant decrease in ABR threshold shift (P0.05).@*CONCLUSION@#The results indicate that the treatment with L-NAC may effectively protect against the styrene-induced hair cells damage and preferably reduce the number of apoptotic OHCs.

ABSTRACT

ObjectiveTo investigate the etiology, clinical characteristics, diagnosis and treatment of regional portal hypertension caused by left upper abdominal malignant tumors.MethodsFrom January 2006 to December 2009, a total of 8 patients presenting regional portal hypertension were treated at our hospital, whose clinical data were analyzed retrospectively. ResultsPancreatic tumors (5/8) and retroperitoneal tumors(3/8)were the primary etiology,andthe main symptoms included upper gastrointestinal bleeding and irregular left upper abdominal pain.Isolated gastric varices were the most distinct clinical features. All patients underwent multi-visceral resection including pancreatic body and tail and spleen. Tumor involved stomach, left kidney, left adrenal and splenic flexure of colon were also removed en bloc. During the follow-up period there was no recurrent upper gastrointestinal bleeding, one patient died and two patients developed metastasis or tumor local recurrence.ConclusionRegional portal hypertension caused by malignant tumor was relatively rare,aggressive resection of multi-viscera combined with devascularization was an effective therapy.

ABSTRACT

Objective To explore the clinical effect and safety of bisoprolol combined with methimazole in treatment of hyperthyroidism.Methods 100 cases with hyperthyroidism were randomly divided into control group (methimazole treatment)50 case.$and treatment group(bisoprolol combined with methimazole treatment)50 cases.The therapeutic effect,serum levels of FT3,FT4 and s-TSH were observed.Results The total effective rate in treatment group 92.0% was significantly higher than 78.0%in control group(χ2=3.798,P<0.05);The contents of TT3,TT4,FT3,FT4 after treatment were lower than before treatment(t=2.245,2.315,2.411,all P<0.05);The contents of S-TSH was higher than before treatment(t=2.335,P<0.05);Hyperthyroidism exponential integral in treatment group decreased significantly than the control group(t=2.312,P<0.05).Conclusion The bisoprolol combined with methimazole treatment could efficiently improve clinical symptoms,raise therapeutic effect in patients with hyperthyroidism.

ABSTRACT

Objective To study Barrett's esophageal consistency with its histological findings,which were found with magnifying chromoendo6copy,and to advance the endoscopic diagnosis of Barrett's esophagus. Methods The patients with BE were diagnosed by endoscopy and histology, and their histological findings and endoscopic appearances of a total of 67 patients with BE were observed. Results Endoscopic appearances type of BE showed island (28 cases) ,tongue(7 cases) ,and circum ference(32 cases). Three types of BE under magnifying chromoendoscopy,and in magnifying endoscope group,type Ⅲ which were found with magnifying endoscope were verified intestinalisation epithelium pat ho-type, compared with type I and type Ⅱ,the discrepancy of them had statistical significance(P<0.01). Conclusion Magnifying chromoendoscopy had a relatively better consistency in the diagnosis of Barrett's esophagus with histological findings.

ABSTRACT

ObjectiveTo investigate the clinical characteristics,diagnosis,treatment and prognosis of limited stage primary esophageal small cell carcinoma (PESC).MethodsClinical data was retrospectively analyzed for 42 patients with pathologically confirmed PESCs who underwent transthoracic esophagectomy with lymphadenectomy from Nov.1990 to Dec.2010 at the Cancer Hospital of Shantou University Medical College.The survival analysis was performed using Kaplan-Meier method.ResultsThe clinical symptoms,imaging and endoscopic features of PESC were similar to those of esophageal squamous cell carcinoma (ESCC).Of the 26 cases that received pre-operative endoscopic biopsy,only five cases were diagnosed as PESC,while the other 21 cases were misdiagnosed as ESCC.The mean follow-up time of this series was 25.3 months (0-123 months).34 patients died of the disease during the follow-up;7 were still alive and 1 was lost.The median survival time (MST) of the 41 patients was 13.0 months (95 % confidence interval 6.3-19.7),and the 6-,12-,24-,36-,and 60-month overall survival rates (OS) were 78.6 %,57.5 %,30.8 %,23.7 %,10.5 %,respectively.ConclusionPESC is a rare disease with poor prognosis,and is prone to be misdiagnosed by endoscopic biopsy.Currently no standard treatment has been established.

OBJECTIVE@#To observe the changes of filamentous-actin (F-actin) during process of apoptosis and necrosis in hair cells, and investigate the early mark for hair cells pathology following noise exposure.@*METHOD@#Twenty young SD rats were used in present study. The animals were randomly assigned into two groups, 10 animals were exposed noise and the remaining without exposure to noise served as normal control. The animals were exposed to a narrow band noise at 110 dB SPL for 8 hours. Immediately after exposure to continuous nose, the animals were exposed to 75 pairs of impulse noise at 155 dB SPL. The auditory brainstem response (ABR) thrush olds of both ears elicited by tone bursts at 5, 10 and 20 kHz were measured before, immediately and 2-week after the noise exposure. Animals were sacrificed and cochleae were quickly removed from the skull. Following fixation, whole specimens comprising the basilar membrane with Corti organ were separated from the modiolus. The organs of Corti were double stained with propidium iodide (PI), a DNA intercalating fluorescent probe used to visualize the morphologic viability of hair cell nuclei, and FITC-labeled phalloidin, a F-actin intercalating fluorescent probe used to visualize the morphologic viability of cuticular plate and the stereocilia in the hair cells. Each organ of Corti was thoroughly examined using fluorescence microscopy.@*RESULT@#The animals of test group exhibited a significant elevation of ABR threshold shifts at all tested frequencies after noise exposure immediately and 2-week. There was no detectable F-actin change in both the corresponding cuticular plates and the stereocilia in the OHCs undergoing necrosis. OHCs with slightly increased PI fluorescence and a relatively normal nuclear morphology exhibited normal level of F-actin activity. In the OHCs with mild nuclear condensation (about 3/4 of the normal size) exhibited normal or a slight increased F-actin fluorescence staining. In the OHCs with moderate or severe nuclear condensation (about 1/2 or 1/4 of the normal size), the reduction in F-actin staining was observed. Finally, in the areas where no detectable nuclear staining was present, the reduction in F-actin staining was more severely or absent.@*CONCLUSION@#These results suggest that compare with the change of F-actin, nuclear morphology and staining is the early mark for necrotic and apoptotic hair cells pathology following noise exposure. A biphasic change in F-actin may exist during apoptosis, an initial polymerization and a subsequent depolymerization, but F-actin may be not a necessary component of the process of necrosis in the noise-damaged OHCs.

ABSTRACT

To investigate the effects of Tacrolimus ointment on patients with erosive oral lichen planus. A total of 62 cases were collected and divided into two groups randomly: 31 cases were treated with Tacrolimus ointment (treatment group), and 31 cases with clobetasol propionate ointment (control group). There was no significant difference between two groups of the effective rate and side-effect occurrence during the therapy, and relapse rate within six months after treatment. The results of the present work demonstrate that Tacrolimus ointment is an effective therapy for oral lichen planus.

OBJECTIVE To investigate the produce of extended-spectrum ?-lactamases(ESBLs) and the presence of genotype of the ?-lactamases-encoding genes in Klebsiella pneumoniae isolated from the 98th Hospital of PLA,Huzhou,Zhejiang Province,China.METHODS Twenty-five strains of K.pneumoniae were isolated from the inpatients between Sep 2005 and Apr 2006.ESBLs were tested by phenotypic confirmatory tests recommended by CLSI.Twenty-one kinds of ?-lactamases genes of blaTEM,blaSHV,blaLEN,blaOKP,blaCTX-M-1 group,blaCTX-M-2 group,blaCTX-M-9 group,blaOXA-1 group,blaOXA-2 group,blaOXA-10 group,blCARB,blaPER,blaVEB,blaGES,blaLAP,blaDHA,blaACT/MIR,blaCMY/MOX,blaFOX,blaCMY/LAT,and blaACC were analyzed by PCR and verified by DNA sequencing.RESULTS In 25 strains of K.pneumoniae,the positive,negative,and "uncertainty" rates of ESBLs were 56.0%,20.0%,and 24.0%,respectively.The positive rate of genes of blaTEM,blaSHV,blaCTX-M-1 group,blaOXA-10 group,blaLAP,and blaDHA were 80.0%,4.0%,4.0%,80.0%,4.0% and 32.0%,respectively.The 15 kinds of rest genes were all tested negative.The total positive rate of 21 kinds of ?-lactamases gene was 92.0%.Among them,the blaLAP-2 gene sequence of the HZ12593 strain has been registered in GenBank(GenBank Accession Number: EU529981).CONCLUSIONS There are higher rate of ESBLs-producing strains in K.pneumoniae isolated from the inpatients,and at least 6 kinds of ?-lactamases gene existed.Both genes of blaTEM and blaOXA-10 group are the most common genotypes.Carring blaDHA Gene may influence the result of phenotypic confirmatory test for ESBLs in K.pneumoniae.
